参考用途:本质控品由河南省工业微生物菌种工程技术研究中心研制,为毛霉属灭活悬液,通过添加稳定剂、数字PCR定值后分装而得,可用于方法建立、工作标准赋值、方法确认与评价、产品质量控制等。
形态特性:小型丝状真菌,在综合 PDA 培养基上菌落明显,初期菌丝白色,后期变灰褐色,培养基背面淡黄色
培养基:马铃薯浸粉:10.0g,葡萄糖:20.0g,KH2PO4:3.0g,MgSO4·7H2O:1.5g,硫胺素:0.008g,琼脂:15.0g,pH:6.0±0.2(25℃)
培养条件:28℃,3-5天,好氧
培养方法:①准备 1 支含有 5~10mL 液体培养基的试管和 2 个平板;②安全柜中打开,用酒精灯灼烧顶部,后迅速滴上无菌水使之破裂,随后用镊子将其敲碎;③吸取 0.5mL 液体培养基打入冻干管,充分溶解后重新打回液体试管中,混匀;④吸取 0.2mL 菌悬液打入平板中,涂布均匀,重复两次获得两个平板;⑤将液体试管和平板全部置于上述培养条件下培养,菌种长出即可使用。
