食品安全国家标准 植物源性食品中331种农药及其代谢物残留量的测定 液相色谱-质谱联用法

适用范围:适用于植物源性食品(蔬菜、水果、谷物等)中331种农药残留的定性和定量分析,涵盖有机磷、氨基甲酸酯、三唑类等常见农药类别。

核心检测方法

1. 样品前处理:QuEChERS方法(乙腈提取,盐包脱水净化)
2. 仪器分析:液相色谱-三重四极杆质谱联用(LC-MS/MS)
3. 色谱条件:C18色谱柱,乙腈-0.1%甲酸水溶液梯度洗脱
4. 质谱模式:多反应监测模式(MRM),保留时间和离子对双重定性

检出限与定量限

检出限(LOD):0.001~0.01 mg/kg(根据不同农药特性)
定量限(LOQ):0.005~0.05 mg/kg,乙腈中农药混标溶液需稀释至标准曲线最低点浓度(通常0.5~5 ng/mL)

质控样品要求

1. 每批次样品需带基质匹配标准曲线(空白样品基质配制)
2. 加标回收率控制范围70%~120%
3. 每20个样品插入1个空白对照和1个加标质控样(加标浓度LOQ水平)
4. 连续分析时每24小时进行仪器校准

关键实验步骤

1. 标准溶液配制:乙腈中51种混标(10μg/mL)用乙腈逐级稀释为6点系列标准溶液
2. 样品提取:10g样品+10mL乙腈振荡提取,MgSO₄/NaCl盐包脱水
3. 净化:取上清液经PSA/MgSO₄净化管离心
4. 上机分析:取净化液过0.22μm滤膜,LC-MS/MS进样(进样量5μL)

特别说明

1. 乙腈中混标溶液需-18℃避光保存,开封后有效期3个月
2. 特别注意基质效应:谷物类样品需增加石墨化碳黑(GCB)净化步骤
3. 部分农药(如涕灭威亚砜)需在提取后立即加入0.1%甲酸稳定
4. 当检出目标物时,需通过离子丰度比(±30%)和保留时间(±0.1min)双重确认
